چکیده

Cerebral Autosomal Dominant Arteriopathy with Subcortical Infarcts and Leukoencephalopathy (CADASIL) is considered a model of pure vascular dementia (VD) because it occurs in young adults unlikely to have concomitant age and Alzheimer's Disease-(AD-) related pathology. CSF levels of β-amyloid 1-42 (Aβ42), total tau protein (t-tau), and phosphorylated tau-protein (p-tau), well accepted biomarkers of AD, were evaluated in 10 CADASIL patients, 22 AD patients, and 17 healthy age-matched subjects. Innotest β-amyloid 1-42, Innotest hTAU-Ag, and Innotest Phospho-tau 181p sandwich enzyme-linked immunoassay were used to determine CSF biomarkers levels. A case-control statistical analysis was carried out. CSF Aβ42 levels were significantly lower in CADASIL patients and considerable overlap with AD whereas t-tau and p-tau levels were normal and significantly different with respect to AD. A significant altered CSF biomarkers profile in a pure VD supports the use of CSF Aβ42, t-tau, and p-tau levels in the differential diagnosis of VD and AD.

P 62: Markers of Neuroinflammation Related to Alzheimer\'s Disease Pathology in the Elderly

Alzheimer Disease (AD) is a neurodegenerative disorder and the most common form of dementia. Increasing evidence suggests that Alzheimer's disease pathogenesis is not restricted to the neuronal compartment, but includes strong interactions with immunological mechanisms in the brain. In vitro and animal studies have linked neuroinflammation to Alzheimer's disease (AD) pathology. Studies on marke...

Cognition in CADASIL.

CADASIL is an early onset small vessel disease and genetic variant of pure subcortical ischemic vascular dementia (SIVD). The condition has been invaluable in defining the profile and neuroimaging correlates of cognitive deficits in pure SIVD. The recent completion of a randomized trial in cognitively impaired CADASIL patients has illustrated the feasibility of targeted therapeutic trials in na...

Should CSF Biomarkers Support a Routine Analysis for Early Diagnosis of Alzheimer's Disease?

In view of the availability of drugs that may slow or even halt the progression of Alzheimer’s disease (AD), the possibility to routinely discriminate individuals carrying incipient AD is utmost important. The CSF biomarkers beta-amyloid and tau are increasingly used in several countries in Europe for their high diagnostic accuracy in detecting early or even prodromal AD. Up till now, thousands...

Neurochemical biomarkers in the differential diagnosis of movement disorders.

In recent years, the neurochemical analysis of neuronal proteins in cerebrospinal fluid (CSF) has become increasingly accepted for the diagnosis of neurodegenerative dementia diseases such as Alzheimer's disease and Creutzfeldt-Jakob disease. CSF surrounds the central nervous system, and in the composition of CSF proteins one finds brain-specific proteins that are prioritized from blood-derived...

Cerebrospinal fluid markers for differential dementia diagnosis in a large memory clinic cohort.

OBJECTIVE To determine how amyloid β 42 (Aβ42), total tau (t-tau), and phosphorylated tau (p-tau) levels in CSF behave in a large cohort of patients with different types of dementia. METHODS Baseline CSF was collected from 512 patients with Alzheimer disease (AD) and 272 patients with other types of dementia (OD), 135 patients with a psychiatric disorder (PSY), and 275 patients with subjectiv...
